You have to be straight forward with him. Don't play games or beat around the bush.
Just tell him: "Listen, I'm not interested. You need to relax and go back to being my friend."
If he persists let him know that it's friend or nothing. Stay firm and be honest. If he's being very aggressive you may have to face the idea that your friendship has run its course and remember it for the wonderful way it was. Sometimes feelings go too far to ever turn back, so prepare yourself.
But if you aren't honest with him, you can only blame yourself for the uncomfortability in your relationship.

Back off from what?
Back off from making a pass at you? Say, "Sorry, I'm into women".
Back off from making a pass at your wife? Say, "She's with me."
Back off from standing too close with bad breath? Say, "Hey... here's a tictac."
Back off from getting too emotional about leaves being dumped in his backyard? Say, "Back off pal... I'll get the kids to clean it up."
Back off from being too drunk and looking for a fight? You can't tell him anything, because he's drunk. But because he's drunk, then if you're not, you ask for some help, and then because two people can overpower almost anyone, you take him down, and tie him up with his shoelaces... but that's telling him to back off.
Mostly, you might find yourself wanting to tell someone to back off if you've done something wrong, and they are going way over the top getting mad at you about it, in which case, about the only thing you can say, "Okay, I get it. I'm sorry. I'll fix that. Please back off."
