Without a doubt, let them know. As far as I'm concerned, grieving for a cat is not as hard as not knowing what happened to it. Did it get sick, hurt, is it hungry, or did it got snagged up by an eagle.
I would not only confess and apologize, I would offer to replace the cat (if that seemed appropriate at the time) or in anyway be of assistance to them.

I actually had this happen to me. My neighbor ran over my cat one night and they never told me. After time passed and we had had a tiff sort of they were bragging about how they killed my cat. It hurt more than I can say. I would say yes, you should definitely tell them and apologize.

Did they then eat it, after running it over ? I would be surprised if they did not, especially the way you said they were gloating.Please check if they flap around at night, because, what you just described, is not normal human behaviour. Accidents can happen, they might even decide to not tell you out of fear, shame , sorrow, but bragging later is not a human trait.I would definitely confess immediately, I would not offer to replace it though, A pet is not a commodity to be replaced, there are a lot of emotions involved.
Anyhow, you be careful of your neighbours.
